is she your fiancee or your wife? Your title says one, and your post another.
For sponsorship, you can send it all in at the same time, to save yourself some apart time, but then your fiancee/wife and kids will have to have their medical exam before you send in the forms.
The forms will need to be filled differently if you're married or not. Canada does not have a Fiancee Visa like the US does. They do have a conjugal partner sponsorship, or spousal sponsorship, depending on your situation.
Its an expensive and frustratingly long process, but at least if you change your mind, or are denied for any reason, CIC will refund 75% of your paid fees. US immigration doesnt do that.
