Work permit is the best way, and it's what CIC recommends, it requires labour certification by
HR Canada, which the employer has to complete to show they need you.
Once you have the permit you can adjust status to permanent resident more easily, as you get more points on the skilled worker test if you're already employed in Canada. Plus you can still live and work in Canada while the application is processed.
Bear in mind if you're a US citizen that the US has a tax law that makes working outside the US disadvantageous, have a read of the instructions for IRS Form 2555. As a US citizen you have to file a 1040 forever regardless of where you live, a limit was imposed on the foreign tax credit in 2006 which means if you earn over $85,700 you are subject to dual taxation, although there are living expense deductions you can claim (but they're not that helpful if you live in Canada).
